Vancomycin is a glycopeptide antibiotic isolated from Streptomyces orientalis. It is currently the first choice for the treatment of penicillin-resistant bacteria and those allergic to β-lactam antibiotics. It has been hailed by international antibiotic experts as “the last line of defence for humans against persistent drug-resistant strains”. As public awareness of drug safety increases, the pharmaceutical industry is demanding ever-higher standards of purity for vancomycin. The United States Pharmacopoeia requires a purity of 90% for vancomycin.
The system for producing daptomycin by microbial fermentation is very complex. The fermentation broth contains a large amount of impurities such as unused culture medium, microbial cells and their metabolites, and the presence of these impurities has caused significant difficulties in the separation and purification of daptomycin. Bona Technology uses ceramic membranes, developed and manufactured in-house, to achieve high-precision, high-throughput filtration and clarification of daptomycin fermentation broth, together with control of filtration process parameters to significantly reduce energy consumption and production costs. After the clarified fermentation broth is subjected to a series of processes such as chromatography, it is concentrated by optimising Bona Technology’s superior performance nanofiltration membrane, and then freeze-dried after concentration to a certain degree to obtain a high-purity daptomycin raw material. Instead of traditional organic solvent extraction, chromatography and other processes, it avoids the use of large amounts of solvents, complicated procedures and large equipment investment, and realises a new, efficient and low-cost separation and purification of daptomycin.
